Water, tools and first aid. Oh, and beer.
I like to keep mine as slim as possible but I ususally have:
Water or gatorade
phone
multi-tool
spare derailleur hanger
tube (for a quick-fix on my tubeless setup)
hand pump (no luck with CO2 in the past)
pins for Shimano chain

Just try and prepare for most of the common break downs like tube, chain and loose bolt. I always do water with a scoop of gatorade powder (just enough to tease you), multi tool, 2 tubes, pump (never do CO2!), sram gold link, food (PBJ's work the best since they are light and good energy), key's/cell/wallet because what the point in having a backpack if you have to carry stuff in your pockets still lol.

Good bits of info about the CO2! Hand pump it is...(wouldn't have thought about the spare derailleur hanger..good!)
Pre-fuel'd on vegetable juice and coconut water--drink TONS of water and eat whole fruits right before heading out
Pack dense foods such as almond butter (more nutritious than peanut), homemade hemp seed dried fruit trail mix concoction with pitted dates (save weight! ..ha) ...that's 100% rocket fuel!!!
